All Rights Reserved.): A composite of wild-type Cryptococcus neoformans cells and mutants lacking a GDP-mannose transporter (Gmt1) highlights the difference in the polysaccharide capsule sizes between the two populations. To distinguish the strains, the walls of the wild-type cells were tagged with aminofluorescein (green) before both strains were labeled with anti-capsule antibody (monoclonal antibody F12D2, generously provided by Thomas Kozel) followed by an Alexa Fluor 546-tagged secondary antibody (red); the cells were then mixed. Although all cells were grown under conditions that normally induce formation of large capsules, the mutant capsules remained small, with staining immediately adjacent to the cell wall, while wild-type capsules were markedly more extensive.
